Israeli forces closed on Wednesday Huwwara checkpoint south of Nablus preventing movement of cars on a major north-south West Bank road.
"The settler was severely injured and taken to the hospital", the spokesperson said, adding that troops were searching the area for the assailant.
An Israeli man was shot dead in the West Bank on Tuesday, close to an illegal settlement near Nablus, the army said. Israel's Channel 10 television station said the man sustained multiple shots to his upper body while he was driving in his vehicle.
Israeli President Reuven Rivlin vowed that "security forces will track down the perpetrators and bring them to justice".

Most countries consider as illegal the Israeli settlements built in the territory which Israel captured in the 1967 Middle East war. It annexed East Jerusalem, saying the entire city is its indivisible capital, in a move that was not internationally recognized.
US -led Peace talks between Israel and the Palestinians broke down in 2014. Some 400,000 Israelis live in settlements in the West Bank, which is home to 2.2 million Palestinians.
